چکیده

• Tyrosol acetate and hydroxytyrosol have wide applications both as functional food components nutraceuticals. Construction of tyrosol biosynthetic pathway in E. coli . derived from virgin olive oil olives extract, However, they low bioavailability due to their absorption high metabolism human liver small intestine. Acetylation can effectively improve thus increase potential use the cosmeceutical industries. There is no report on bioproductin so far. Thus, it great significance develop microbial cell factories for achieving or biosynthesis. In this study, a de novo production was constructed Escherichia First, an engineered that allows simple carbon sources established. Four aldehyde reductases were compared, found yeaE best reductase accumulation. Subsequently, extended by further overexpression alcohol acetyltransferase ATF1 conversion acetate. Finally, 4-hydroxyphenylacetate 3-hydroxylase HpaBC.

Biosynthesis of histidine from radioactive acetate and glucose.

Evidence has been presented in a previous paper (2) that formate is the precursor of carbon atom 2 of histidine in yeast and that the imidazole ring of the amino acid is not derived from the corresponding ring of the purine bases. The present paper is concerned with the origin of the 5 carbon chain of histidine. The studies of Edlbacher (3) indicated that histidine catabolism in liver preparati...

Physiological and Molecular Timing of the Glucose to Acetate Transition in Escherichia coli

The glucose-acetate transition in Escherichia coli is a classical model of metabolic adaptation. Here, we describe the dynamics of the molecular processes involved in this metabolic transition, with a particular focus on glucose exhaustion. Although changes in the metabolome were observed before glucose exhaustion, our results point to a massive reshuffling at both the transcriptome and metabol...

Eliminating acetate formation improves citramalate production by metabolically engineered Escherichia coli

BACKGROUND Citramalate, a chemical precursor to the industrially important methacrylic acid (MAA), can be synthesized using Escherichia coli overexpressing citramalate synthase (cimA gene). Deletion of gltA encoding citrate synthase and leuC encoding 3-isopropylmalate dehydratase were critical to achieving high citramalate yields. Acetate is an undesirable by-product potentially formed from pyr...

The utilization of acetate for synthesis in Escherichia coli.

For many years the importance of the Krebs tricarboxylic acid cycle in bacterial metabolism has been contested. Some authors find evidence for the occurrence of the cycle in certain species of bacteria; in other cases, results are interpreted as proving that the cycle does not operate (see Ajl for a recent review (1)). In most of these studies, as indeed with those on other organisms, the empha...

Inhibition of de novo purine biosynthesis and interconversion by 6-methylpurine in Escherichia coli.

The inhibition of Escherichia coli strain B and strain W-11 by 6-methylpurine depended on the formation of 6-methylpurine ribonucleotide by the action of adenine phosphoribosyltransferase (AMP: pyrophosphate phosphoribosyltransferase, EC 2.4.2.7). 6-Methylpurine ribonucleotide inhibited the de novo synthesis of purines, presumably via pseudofeedback inhibition of phosphoribosylpyrophosphate ami...
